Can you mix AMD and Nvidia cards on the same PC (when mining of course)?

Also what watt PSU would I need for the (3) 1080tis and the (1) RX480
Running the 1080tis at 80% load only.

I have an EVGA GQ 850 Watt 80 Plus Gold

The Mobo is a ASUS PRIME Z270-AR LGA 1151 and supports Quad SLI

For the power question, I believe it may come down to the power consumption of the RX480.  From what I understand, you may want to stay @ 80% load for PSU- 680W for 850W.  I would assume your current load is under 680W- 3 x 1080 ti @ 80%= 250W x 0.8 x 3= 600W; if so appears this leaves 80W to not exceed 80% threshold.  Not sure if 80W for the RX480 will yield the mining result you are looking for.  If it does, you are good; if not, may want to consider a 1000W PSU or possibly higher for future expansion- measure twice cut once as opposed to measuring once and cutting twice.
