You certainly can! You can add the gift card during checkout using the Amazon Prime Now app or you can add it before you check out. If you add it before you checkout, the gift card balance will appear as a payment option during the checkout process.

This is not necessarily correct, to my knowledge Amazon Prime is based on your account rather than the payment method/gift card.  Regardless, you can certainly use the gift card to pay for Prime!
