The maximum amount per transaction is 999 BTC so it doesnt matter, it depends on you how fast you wanna have your bitcoins anonymized, typically it takes up to 6 hours, but you can also divide your btc into small transaction but it will take too long because every single anonymization takes up to 6 hours so it will be faster if you do 5 BTC at once.
