No. But there are disadvantages: A LN wallet is always a hot wallet, and large funds should be kept safe in cold wallets.
You could probably open a very large channel with someone (you may need to patch the maximum amount in the software) and then send him 35 Bitcoin directly, but it really defeats the purpose of LN. LN's advantage is being able to make many transactions at low fee after opening an channel (which requires an on-chain transaction).
