First you need have some coins...than setup in options how many coins you want anonymised and how many rounds....more rounds cost you more...and start mixing...it will take some time but you will see the progress of it....once it get 100% you can send the coins anonymously by ticking the Darksend

No. It was anonymous at step 3, but when you buy BTC on exchange, the exchange can link the BTC you withdraw to your exchange account.
If you want to buy something anonymously, do steps 1-3 and buy it with DASH.

However, how to enable darksend?
In my wallet, it shows as "disabled", although i have set "darksend rounds" to "3".
(i have 0 dash, if it matters ?)

You can sent normal DASH (not anonymized, ie: without darksend) or anonymized DASH, you have to darksend them before.Check my first post I edit it with a link for more information.
Yes, is ahead of time mixing so is not transaction based privacy, you decide how much money you want to keep on your private balance and the level of privacy you want to have and then the wallet mixes the coins for you up to 16 rounds, and then your private balance is there ready for you to spend at any time, without you having to wait for peers or remix you just send them directly as any Bitcoin transaction. It is really cool because it provides a high level of privacy while keeping the same mechanics we are used to with BTC.

You have to click the "Start Darksend Mixing" when you have some coins in your wallet. This will anonymized your coins, and then they will be ready to send when ever you want.

and after i received their dash, i click the "Start darksend mixing", the mixing process done, and all of my dash become anonymous, and i can spend my dash?

You have to initialize the process to mix your coins. After your coins are mixed, you send like any other BTC tx with the DS box checked.
