I am afraid that there is no good way to do that, i think you can count amount of wallets... But i am not sure about that either, as probably wallet owners will not want to give this info

or people just can have a few wallets, all in all it is impossible and i doubt that anyone will be able to count the users of bitcoins

we will most likely never know, but you can see that the numbers of users are growing since the transaction volume is increasing, and blocks are nearly always full. that's a good indication of how the actual usage is growing.
